Isolation and Characterization of Epidermal DNA and RNA from Guinea Pig Skin

1971

DNA and RNA were isolated from mammalian epidermis in a relatively small scale procedure. The high purity and native state of the DNA isolated is reflected by its molar absorptivity E (P), its thermal hyperchromicity and its hyperchromicity upon DNase treatment and by its sedimentation profile as well as by its profile in a cesium chloride density gradient. The very low content of protein and RNA, as well as the data of DNA determination, indicate that this method permits the isolation of a highly purified product. This is further substantiated by the determination of UV absorption spectra and by analysis of the base composition. The mammalian skin DNA showed the following properties: Mol. w…

The molecular heterogeneity of hemocyanin: Structural and functional properties of the 4×6-meric protein of Upogebia pusilla (Crustacea)

2006

The structural properties of the hemocyanin isolated from the Mediterranean mud shrimp, Upogebia pusilla (Decapoda: Thalassinidea), were investigated. Our intent was to make use of the U. pusilla case to perform a structural comparison between crustacean and chelicerate 4×6-meric hemocyanins. The thalassinidean hemocyanin appears similar in size but different in structural organization compared to the chelicerate 4×6-mer. Ultracentrifuge analyses on the purified protein revealed a sedimentation coefficient of 39S, typical of 4×6 hemocyanins. Electron micrographs are in agreement with a model in which four 2×6-meric building blocks are arranged in a tetrahedron-like quaternary structure and …

ISOLATION OF RAT LIVER EPOXIDE HYDRATASE: PROPERTIES AND SUBSTRATE SPECIFICITY OF THE PURE ENZYME

1977

ABSTRACT The microsomal enzyme epoxide hydratase has been purified to homogeneity as judged by electrophoretical, ultracentrifugal and immunological criteria and by C- and N-terminal analysis. The preparation procedure consisted of solubilisation using the non-ionic detergent cutscum, (NH4)2SO4 precipitation, ion-exchange chromatography on DEAE-cellulose and cellulose phosphate and hydrophobic chromatography on butyl-sepharose. The product was detergent-free, had a relatively high content of hydrophobic amino acids and tended to aggregate in aqueous solutions. The protein had a minimum molecular weight of 49,000 ± 500 with a sedimentation coefficient of S20w≃ 3. Antibodies raised against th…

Developmental changes and acetylcholinesterase activity in the metamorphosing brain ofTenebrio molitor: Correlation to ecdysteroid titers

1994

The brain of Tenebrio molitor exhibited marked fluctuations in acetylcholinesterase (AChE) activity throughout metamorphosis. This was true AChE activity, since it was inhibited by high substrate concentrations and by 10 μM of the specific AChE inhibitor BW284C51 [(1,5-bis'4-allyldimethylammoniumphenyl)-pentan-3-one dibromide] but not by iso-OMPA (tetraisopropylpyrophosphoramide), a cholinesterase (but not AChE) inhibitor. The histochemical AChE activity was localized in the neuropile and the nuclear envelope of neurons and glial cells. The enzyme extracted from brains with 1% Triton X-100 and 1 M NaCl sedimented as a single peak in a sucrose density gradient, with a sedimentation coefficie…
